13.Teadustöö põhisuunad a) Characterization of binding sites and interaction to neuropeptides of serotonin reuptake inhibitors citalopram, fluoxetine and other antidepressants, projektijuht prof. L. Allikmets.
Projekti põhieesmärgiks oli iseloomustada selektiivsete serotoniini tagasihaarde inhibiitorite (SSRI) sidumiskohtade neurokeemilisi omadusi ning SSRIde võimalikke interaktsioone neuropeptiididega. Katsete tulemusel selgus, et SSRIdel ei ole olulist neurokeemilist interaktsiooni koletsüstokiniinergilise närviülekandega, kuid selline seos võib ilmneda käitumuslikul tasandil kaudsete mehhanismide vahendusel suurenenud ärevusnivoo tingimustes.
b) The mechanism of affective disorders and the kinetics and mechanism of action of the novel antidepressants, projektijuht prof. L. Allikmets. Projekti raames uuriti põhjalikult erinevate serotoniini retseptorite osa anitidepressantide toimemehhanismis, samuti teiste psühhotroopsete ainete kaudset mõju ning füsioloogilisi faktoreid, mis mõjustavad antidepressantide toimete avaldumist. Katsete tulemused näitasid, et serotoniini 5-HT2A/2C retseptorid võivad olla haaratud antidepressantide ja ka antipsühhootikumide toimetes.
c) Elucidation of the mechanism of anxiogenic/anxiolytic action of antidepressants, projektijuht prof. L. Allikmets. Projekt on tihedalt seotud eelneva projektiga, eelnevale lisaks uuriti põhjalikult 5-HT1A jt serotoniiniretseptorite osa neurokeemilistes ja käitumiskatsetes rõhuasetusega emotsionaalsel käitumisele. Tulemuste alusel võib väita, et serotoniinergilise närviülekande osa on antidepressantide poolt vahendatud emotsionaalse käitumise efektides kompleksne ning ei ole võimalik välja tuua selgelt hirmu tekitavat/vähendavat serotoniini retseptoralatüüpi.
d) Mechanism of action of second generation antipsychotics: interaction with glutamatergic and serotoninergic systems, projektijuht prof. L. Allikmets. Projekt on suunatud uute, atüüpiliste antipsühhootikumide toimemehhanismide täpsustamisele. Selgitati põhiliselt dopamiinergilise närviülekande ergastumisest kui kaootilise käitumise mudelist tulenevate fenomenide neurobioloogiat ja nende mõjutamist uute, atüüpiliste antipsühhootikumidega.
e) Role of accumbal dopamine in molecular mechanisms of ethanol dependence, projektijuhid prof. Liisa Ahtee ja prof. T. Petteri Piepponen, HÜ Farmaatsia teaduskonna Farmakoloogia ja Toksikoloogia osakond. Töö pôhitulemuseks oli, et dopamiini vabanemine N. Accumbensis on osaliselt sôltuv etanooli manustamise viisist.
f) Sleep in aging women (EU project QLK6-CT-2000-00499), projekti koordinaator dotsent, Soome Akadeemia vanemteadur Tarja Porkka-Heiskanen (HÜ Biomeditsiini Instituut, Füsioloogia osakond), alaprojekt Non-REM sleep and basal forebrain GABA levels in young and aged female rats.

13.Current research program a) Characterization of binding sites and interaction to neuropeptides of serotonin reuptake inhibitors citalopram, fluoxetine and other antidepressants, PI Prof. L. Allikmets.
The aim of the project was to characterize the binding neurochemical characteristics and interaction with neuropeptides of the selective serotonin reuptake inhibitors (SSRI). The results revealed that the SSRIs do not have a robust interaction with the cholecystokinetic neurotransmission, while such an association is possible on the behavioral level under circumstances of increased anxiety.
b) The mechanism of affective disorders and the kinetics and mechanism of action of the novel antidepressants, PI Prof. L. Allikmets. The project dealt with the elucidation of the role of serotonin receptors in the mechanism of action of antidepressants, as well as psychopharmacological and physiological factors influencing the effects of antidepressants. We found that the serotonin 5-HT2A/2C receptors may be implicated in the mechanism of action of antidepressants and othe psychotropics.
c) Elucidation of the mechanism of anxiogenic/anxiolytic action of antidepressants, PI Prof. L. Allikmets. The project was closely associated with the previous one, in addition the role of serotonin 5-HT1A et al. receptors on the emotional and motivational behaviour was studied. It was found that the serotonin-receptor-mediated emotional impairment is a complex phenomenon without a clear "anxiogenic" 5-HT receptor subtype.
d) Mechanism of action of second generation antipsychotics: interaction with glutamatergic and serotoninergic systems, PI Prof. L. Allikmets. The project was focused on the investigation of the mechanism of action of the novel, second generation antipsychotics. The role of the dopaminergic neurotransmission in the context of novel antipsychotics in the mediation of frantic behaviour was studied.
e) Role of accumbal dopamine in molecular mechanisms of ethanol dependence, PI Prof. Liisa Ahtee and Prof. T. Petteri Piepponen, University of Helsinki, Department of Pharmacy, Division of Pharmacology and Toxicology. The main result of the study was that the release of dopamine in Nucleus Accumbens is partly dependent on the route of ethanol administration.
