description: | OBJECTIVES 1. Construction of delivery system to tag probiotically active L. fermentum ME-3 and other lactobacilli by molecula cloning. This gives the possibility to: 1.1. detect tagged lactobacilli qualitatively in microflora; 1.2. evaluate lactobacilli quantitatively in mouse models. 2. Evaluation of probiotic activity of the DNA-tagged L. fermentum ME-3 and other lactobacilli to the urophatogenic E. coli in the experimental long-term ascending urinary tract mouse model (impact of lactobacilli in urinary tract) and streptomycin treated mouse model (impact of lactobacilli in gastrointestinal tract). POSSIBLE RESULTS 1. According to mouse model results the strain of lactobacilli will be selected which will diminish the pathologic alterations in urinary tract and is also antagonistic against uropathogenic E. coli in vivo. 2. The mechanism of probiotic activity against urophatogenes will be determined. 3. It is possible to use results of experimental investigations in clinical trials in the future. 4. The results of molecular cloning in lactobacilli will open advanced research in the field of interaction of lactobacilli and host. 5. Technology of molecular cloning can be used to construct lactobacilli with medical and dairy importance. |
