description: In this project we are going to investigate gene expression in neural stem cells and in vitro differentiated neurons and glial cells. We are planning to create 2D- electrophoresis databases for expressed gene products (proteins) to get the possibilities for analyzing changes taking place in the cells in normal conditions and due to genetic defects and transfection of foreign genes. We plan to do a comparative study of the transcription factor gene expression both in normal stem cells isolated from different sources and in stem cells from genetically abnormal fetuses and from sphereforming cells from neural tumours before and after differentiation, and also after transfection of foreign genes. In transfection we are using DNA reparation and replication factor genes to study the stability of their integration and the influence of their overexpression (if taking place) to the expression of transcription factor genes and to the proteome of the cells as a whole.We also plan to generate for this work new neural stem cell lines from brain tissue both of spontaneously aborted fetuses and fetuses removed due to medical indications (including fetuses with prenatally diagnosed defects). In this work the cell lines will be generated only for scientific purposes not for the therapy.

Expected results: The 2D- electrophoresis databases for the gene expression (proteomics) in neural stem cells and in vitro differentiated neural cells will be created. The expression patterns of transcription factor genes (>50) will be established both for normal and genetically defective in vitro growing neural cells. It is expected to get new knowledge how this pattern will change by transfection of foreign genes ( in this work DNA replication and reparation factor genes). New neural stem cell lines will be generated and used for this work.
