description: Tissue factor (TF) is a small transmembrane glycoprotein that activates the blood coagulation protease cascade. When this protein is expressed on the surface of cells that are exposed to blood, it activates blood coagulation and can cause coagulopathies in different pathologic conditions. The present study focuses on the clinically important pathologic expression of tissue factor (TF) in myeloid/leukaemic cells. Using the model system of human cell lines, we plan to investigate the mechanisms of TF expression in acute promyelocytic leukaemia (APL) cells and study the retinoid-induced regulation in APL and other cells. (1) Based on our previous knowledge, we plan to identify transcription factor binding complexes responsible for contsitutive TF expression in APL cells. (2) We intend to study the role of APL-specific oncogene PML/RARalpha in the retinoic acid (RA)-induced suppression of TF. (3) We intend to investigate the transcriptional mechanisms involved in RA-induced suppression of TF in the cells that does not express PML/RARalpha fusion protein.
This project helps to reveal the exact mechanisms of TF suppression in different leukaemic cell lines and may have implications for the treatment of leukaemia- associated thrombotic complications.
