description: A number of protein factors involved in mitochondrial DNA metabolism in yeast Saccharomyces cerevisiae have been found. However, the overall replication scheme, and mechanism of replication initiation are still missing for this important model organism. This project has a goal to understand some aspects of DNA metabolism in yeast mitochondria. We would like to analyze the role of conserved ori-rep structures and their potential interactions with protein factors to get a deeper insight of processes leading probably to DNA synthesis initiation in wt mitochondrial DNA. We would also like to analyze replication and recombination intermediates of wt and rho- mitochondrial DNA using mostly 2D electrophoresis. We will also focus on nucleoid proteins in various genetic background and study their involvment in DNA participation process and in regulation of mitochondrial genome copy number. Finally we will analyze mitochondrial DNA copy number control by the mitochondrial dNTP pool. All these deeply integrated questions are important aspects of processes that guarantee faithful propagation of mitochondrial genetic material
