description: | The aim of our project is to observe association of single nucleotide polymorphisms in the IL-10, IL-19 and IL-20 genes with the susceptibility, severity and course of different forms of psoriasis. We will study how different IL-10, IL-19 and IL-20 haplotypes correlate with cytokines production in healthy controls and in patients with psoriasis and investigate the functional significance of various haplotypes to susceptibility, severity and course of psoriasis. For detection of IL-10 SNP-s we apply ARMS-PCR method. Described method has been widely used for screening of multiple SNP-s and recently description for IL-10 SNP-s has been published. We will use the same method for IL-19 and IL-20 polymorphism genotyping. For determination of cytokines expression we will perform mRNA RT-PCR analyzes. This will be the first time to detect so extensively polymorphic variants of IL-10 promoter region in psoriasis.There is no also information about the allele distributions of IL-19 and IL-20 SNPs in psoriasis.Genotypic and cytokine mRNA expression analyses may enable to understand the molecular basis of the involvement of IL-10, IL-19 and IL-20 in the genetic susceptibility to psoriasis and the inter-individual variability of these cytokines quantitative production. |
