description: | We have been cloned and characterized a new human plexin, plexin-B3 (PLXN-B3, GeneBank acc.no. AF149019), which is located at X-chromosome (Xq28) in the intimate vicinity of the L1CAM. It appears that molecular interactions between semaphorins and plexins provide a basis for finely tuned signaling and intercellular cross-talk during organogenesis. We are interested about PLXN-B3 role in the vertebrate development, and about a molecular mechanisms and pathways causing this concrete developmental processes. This is also a main aim of current proposal. Future investigations would allow us: 1. To clarify the role and find regulatory elements (promoter) which influence PLXN-B3 expression in the development of mammlian nervous and other organ systems. 2. To find molecules, which are able to activate PLXN-B3 and molecular partners which bind to PLXN-B3 constituting part of formed active receptor complex. 3. To investigate PLXN-B3 activation mechanisms and molecular pathways, transforming intracellular signals of activated PLXN-B3 to cellular responses. 4. Isolate PLXN-B3 domains and motifs, which are responsible for ligand(s) and downstream effector(s) binding. In conclusion, signaling by semaphorins seems to be a perfect example of ecletic cell-cell communication code, exploited by a veriety of cells and in different instances, from embryo development to adult pathology. From the point of medicine they are important as a protteins involved in the regeneration processes, tumor markers and on the other instance as a potential tumor suppressor proteins. |
