description: The aim of this project is isolation and characterization of L-amino acid oxidases (LAAO) from Vipera lebetina and V. berus venoms. LAAO that has been previously used mainly for synthesis of alpha-ketoacids, has now been of interest due to its effects on different cells: platelets, vascular endothelial cells, tumor cells. The results published about cellular effects are controversial. The LAAO from some venoms (A. halys blomhoffii, N. naja kaouthia) inhibits platelet aggregation, from other venoms induces aggregation. The proposed mechanisms are not clearly defined yet and need further studies. So the purposes of this project are as follows: to study the interactions of isolated LAAOs with platelets and with HUVEC and tumor cells including elucidation of the mechanism of action. We plan to isolate the cDNA encoding LAAO of V. lebetina venom, to sequence it and to translate the cDNA sequence into protein sequence. Primary structure will give essential information about potential interactions of LAAO with other proteins including cell surface receptors.
