description: We have shown that Gata-3-lacZ is expressed in the endocardial cushion, semilunar valves and impulse conducting system of embryonic heart of mouse and Gata-3 "knock-out" embryos have defects of outflow tract and branchial arch arteries. The homozygotes of Gata-3 mutant embryos die between 11-12 dpc and display massive internal bleeding, marked growth retradation, severe deformities of neural system (Pandolfi, et al.,1995).
1.To circumvent early embryonic lethality Gata-3 will be disrupted specifically in the heart by using cre-loxP technic. Now one can characterise the fenotype of older mutant hearts and estimate the role of Gata-3 in congenital heart diseases. Alternatively the cateholamine treatment will used to get older developmental stages.
2.To characterize changes of cellular apoptosis, proliferation, migration and differensation in the hearts with inactivated Gata-3.
