description: Mutations in the AIRE (autoimmune regulator) gene cause APECED (autoimmune polyendocrinopathy, candidiasis and ectodermal dystrophy or APS-I, OMIM *240300). The disease is an autosomal recessive and characterized by a tissue-specific destructive autoimmune process, affecting principally endocrine organs. Previous experiments have shown that AIRE is expressed in myeloid dendritic cells. However, no AIRE expression has been analyzed in plasmacytoid, indolamine-2,3-dioxygenase (IDO) producing “regulatory” dendritic cells or in Langerhans cells, the dendritic like cell usually located in skin. The working hypothesis is that AIRE is also expressed in other dendritic cell subpopulations and the objective of the research plan is to characterize AIRE expression in these cell populations by quantitative PCR (QPCR) and immunocytochemistry. Another hypothesis is that AIRE is stimulating the production of chemokines in dendritic cells, which are important for chemotaxis of T cells.
