description: Identification of differences in transcription factor profiles between normal and BCC cells would indirectly point out the genes that are abnormally expressed and could be important in BCC development. In the current project we undertake to develop a method that would allow profiling different transcription factors in diseased and normal cells/tissues. The information obtained could elucidate details of complex mechanisms behind skin cancer. We have developed a principle of such method. It is based on the ability of double-stranded oligonucleotides corresponding to consensus sequences of transcription factors to bind specific protein in vitro. Intracellular proteins (transcription factors) that bind these oligonucleotide sequences could be “pulled” out from the cell lysate using previously described interaction. According to our approach the cell lysate will be globally labelled and proteins interacting with consensus sequences will be captured. The identification of these proteins will be performed using mass-spectroscopic methods.
