description: Fatty acid hydroperoxides are known as signalling molecules throughout plant and animal kingdoms. Detailed biomedical studies of their biological effects have been hindered by the absence of efficient methods for their synthesis. Authors of this application have identified in coral Gersemia fruticosa 8-, 9- 11-, 12- and 15-HETE and isolated initial clones of several lipoxygenase-related sequences that allowed to assume that novel positionally specific lipoxygenases are present. The current project is aimed in cloning and heterologous expression of these novel lipoxygenases and elucidation of their structures as well as regio- and stereospecificity of fatty acid dioxygenation. Further aims are the optimisation of expression of 8-, 9-, and 11-lipoxygenases (no rich natural sources available) and of biosynthesis of enantiomerically pure hydroperoxy and hydroxy acids for biochemical and pharmacological studies. Unsaturated hydroperoxy acids serve also as precursors for transformation to unsaturated volatile aldehydes (flavour industry) by hydroperoxide lyases.
