title: Reactive changes and muscle regeneration in human muscle cells of the patients with different muscle diseases
reg no: ETF5499
project type: Estonian Science Foundation research grant
subject: 3. Medical Sciences
status: completed
institution: University of Tartu
head of project: Andres Piirsoo
duration: 01.01.2003 - 31.12.2005
description: The recent project based on the previous investigations of muscle pathology, carried out in the laboratory of cell biology and electron microscopy , Medical Faculty, Tartu University in cooperation with Children Hospital ( Prof. Tiina Talvik) and Department of Pathology of University Hospital.
The collection of biopsies , we have already for investigation, exceed more than 120 cases and every year more than 20 new cases will be added to the investigation
The present project is aimed to investigate the reactive changes, regeneration potential and pathogenesis mechanisms of human skeletal muscle of the patients with different muscle diseases Pathomorphological and clinical data and there correlations from this investigation will form the unic data bank about muscle diseases in Estonia and it will be useful not only for the future muscle research but also for clinical diagnostic.
As the realization of regeneration potency of muscle tissue is one of the topic problem in muscle research according to the hope for cell therapy , it will be investigated the expression of extracellular matrix proteins ( laminins, fibronectin, NCAM etc.), muscle specific proteins ( MyoD, myogenin, Myf5 etc.) and several transcription, replication and reparation factors in sattelite cells and their derivatives.
According to the realistic hope for cell therapy, it investigated the expression on stem cell specific marker proteins expression ( CD34,CD38 etc.) in sattelite cells and in other undifferentiated cells of human muscle.
The result expected are the following:1.it will be formed the unic pathomorphological data bank of the muscle disorders of Estonia and the overview about the pathomorphology of muscle disorders of Estonia will be publiched with cooperation of clinicists. 2.it will be investigated the pathogenetic mechanisms of muscle disorders with different background and at least 2 scientific papers will be published ( one presumably about the genesis of cores and the other about the pathomorphology of mitochondrial myopathies). 3.It will be revealed the regeneration potency, their realization and regulation in pathological muscle. An article about the expression of different proteins in regenerative muscle will be published. 4.it will be revealed the reason of abnormal differentiation in regenerating muscle, the nature of aggregates and deposits and the mechanisms of their formation.
